The Zn fingers domain of PLZF is sufficient for the RAR-PLZF interaction. Full length PLZF and isolated domains of PLZF (BTB/POZ, Acidic, intermediary, Proline-rich, Zn Fingers) were synthesized and labeled by in-vitro coupled transcription/translation and then incubated either with a GST-RARα or with RAR K244A-K262A-Sepharose matrix in the absence or presence of 1 μM atRA. Complexes were then resolved and quantified as in Figure 2.
